How is Pfast different from a full Pfenex program?

Like the name, Pfast is fast. Our advanced Pfenex strain engineering program is historically used for extremely challenging proteins or for identified lead candidates where expression optimization is needed. A full platform screen and scale-up is typically 10 to 12 weeks. Pfast accelerates this process using known high-potential toolbox subsets to quickly drive to the optimal expression strategy, or the final strain itself.

Combinations screened
Up to 1,000 unique expression strains generated by combining an array of hosts with a set of expression vectors, exploring a number of genetic elements with your gene of interest.
A pooled ligation strategy where your gene of interest is combined with up to 200 different vector combinations, and transformed into one host.
Screening
Duplicate samples of each clone in an automated 96 well format.
Serially diluted cell pool grown in an automated 96 well format.
Purification tags necessary?
No, custom enrichment strategies can be developed.
IMAC compatible or other affinity tag; untagged proteins may incur additional cost.
Analysis
Multitude of parameters measured, including titer, activity (ELISA, BLI enzymatic activity), HPLC, LC-MS, and many others.

Program Deliverables:

  • Detailed protein titer and analysis of soluble and insoluble fractions via SDS-CGE or ELISA.

  • Sample material for further testing provided if suitable titers achieved.
  • Strategic recommendations for additional strain improvements, or advancement to 2L scale fermentation.
